More commonly known as fat or fatty acids, lipids, together with DNA, proteins and carbohydrates, are one of the four fundamental classes of macromolecules found in nature (Walt D et al., 2012). One of the most important functions of lipids is acting as the building blocks of cellular membranes (Subramaniam S et al., 2011). Other major functions of lipids include energy storage, insulation, cellular communication and protection (Fahy E et al., 2009). Lipids have applications in the cosmetic, medicine, food industries and in nanotechnology (Bernie Y, 2007). Bligh E. and Dwyer W. first performed the extraction and purification of lipids from biological materials like fish in 1959 (Bligh E and Dwyer W, 1959). Since then many new techniques have been developed and employed in lipid extraction and purification.
The separation and purification of lipids using chromatography presents unique challenges to the scientists as they are non-polar in nature and are non-chromophores. However, with the advent of Evaporative Light Scattering Detection (ELSD), isolation and purification of lipids has become relatively easy. Several scientists have published their research (along with the method data) on purification of lipids using BUCHI Flash/Prep chromatography instruments.
